Glass fermenters are commonly used in microbiology laboratories of research institutes, universities and enterprises. They are suitable for the screening of microbial fermentation medium formulas, optimization of fermentation process parameters, production process and verification of strains. It is an ideal test equipment for precision fermentation. .
With the increasing use of fermenters, knowledge about glass fermenters is increasingly being consulted by customers. Today, we have compiled a summary of the application, use, sterilization and cleaning of glass fermenters.
First, the application field of glass fermenter
1. Use in the food industry: There are three main categories of goods, one is the production of traditional fermented goods, such as beer, fruit wine, vinegar, etc.; the second is the production of food additives; the third is to help solve food problems.
2. Use in the pharmaceutical industry: According to the fermentation engineering skills, developed a variety of drugs, such as human growth hormone, recombinant hepatitis B vaccine, monoclonal antibodies of certain varieties, interleukin-2, anti-hemophilia factor, etc. .
3. Use in the field of environmental science: the strengthening of microorganisms in sewage treatment.
Second, the use of glass fermenters
1. Before using the glass fermenter, the liquid must be cleaned with fresh hot water and then steamed. The material slurry enters the cylinder from the material pipe fixed on the cylinder head, or the cylinder head is opened. The material should not be packed. Too full to prevent the material from being splashed when it is stirred, resulting in unsanitary or loss of environment.
2. Heating method: When heating, the refrigerant inlet pipe must be closed, the remaining refrigerant in the jacket should be placed, the material should be input, the agitator should be turned on, and then the steam valve should be opened.
3. After reaching the required temperature, close the steam valve and turn off the stirrer after 2-3 minutes.
4. Cooling method: Close the steam valve, drain the remaining steam condensate from the jacket, and then open the tank bottom refrigerant valve. Passing the refrigerant through the jacket reduces the temperature of the material in the tank.
5. Insulation: According to the required temperature, start the agitator, adjust the valve, maintain the temperature, (pay attention to the temperature gauge), in order to achieve the purpose of insulation.
Third, the glass fermentation tank sterilization method
"Sterilization" refers to the technical or technological process of killing or removing all living substances in materials and equipment by chemical or physical means. The sterilization can be divided into two types: sterilization and lysing. The former means that although the bacteria are dead, However, the form still exists, while the latter refers to the phenomenon that the cells melt and disappear after the cells are killed.
Commonly used sterilization techniques are generally divided into: physical sterilization and chemical sterilization. Physical sterilization includes moist heat sterilization, dry heat sterilization, radiation sterilization, and filtration sterilization. Chemical sterilization is mainly the use of chemical reagents (such as formaldehyde, phenol, neoprene, peracetic acid, potassium permanganate, etc.) for the sterilization of certain containers or materials and sterile areas.
In the aspect of biological fermentation, the commonly used equipment is mechanical agitation fermentation, and the sterilization effect directly affects whether the fermentation can be carried out normally.
Fourth, the cleaning method of the glass fermenter
Glass fermenters should be carefully cleaned before and after use. Especially when different strains are used for the two cultures before and after, the cleaning and sterilization work should be paid more attention. Any part of the glass fermenter that can be cleaned should be carefully cleaned, otherwise it may become a breeding ground for bacteria. The areas that are easily overlooked and cannot be adequately cleaned are the inside of the nozzle corner and the inside of the sampling tube and the top of the tank.
