Studies have shown that many proteins contain low complexity domains. This domain can not only regulate the "self-aggregation" state of the protein through liquid-liquid phase deformation, but also is the main cause of many major diseases such as Alzheimer's disease, Huntington's disease, and amyotrophic lateral sclerosis.
The Jishanming research group used the fruit fly as a model to conduct long-term and systematic research on the enzymatic properties and biological functions of Drosophila Otu. The study found that the low complexity domain can regulate the liquid-liquid phase transformation of Drosophila Otu protein and play an important role in the Otu "self-aggregation" process and deubiquitination enzyme activity. RNA binds to the low-complexity domain of Otu, thereby promoting Otu's "self-aggregation" and positively regulating its enzyme activity. When Otu expression is disordered or protein aggregation is blocked, Drosophila intestinal barrier function is impaired, immune signals are disordered, and lifespan is shortened.
The study further found that the "self-aggregation" behavior and deubiquitinating enzyme activity of Otu in vivo is a regulatable process. The molecular chaperone Bam interacts with Otu and promotes its "self-aggregation", enhances its deubiquitinating enzyme activity, and assists in the regulation of intestinal immunity and longevity in fruit flies. At the same time, the expression of Bam changes dynamically with age, and its dynamic changes can regulate the activity of Otu, thereby regulating the lifespan of fruit flies.
A large number of studies have reported in the past that abnormal aggregation of proteins can have a negative impact on cells and organisms. The development of this study revealed that the protein aggregation/liquid-liquid phase transformation has a positive biological function. Protein aggregation not only controls its activity to cope with physiological and environmental changes, but also plays an important role in maintaining intestinal homeostasis, individual health and delaying aging. The research results have important implications for people to better understand how protein aggregation regulates its own enzyme activity and individual physiological processes.
