1. Summary
Aflatoxins are toxic metabolites of a class of fungi such as Aspergillus flavus and Aspergillus parasiticus, which are highly carcinogenic and aflatoxin M1 is a metabolite of aflatoxin B1 . The milk produced by the cows containing the aflatoxin B1 feed contains aflatoxin M1 . Since aflatoxin M 1 is quite stable and cannot be killed by pasteurization, it is necessary to strictly test the content of aflatoxin M 1 in milk and products. China, the United States, Japan, Korea, and the International Codex Alimentarius ( CAC) ) The aflatoxin M 1 in milk should not exceed 0.5 ng/ mL (ppb) . The iCheck® Aflatoxin M 1 Quantitative Fast Check Card allows rapid and quantitative determination of aflatoxin M 1 in the 0-2 ppb range of raw milk and bagged pure milk.

3. Equipment required for testing
1) 200μL, 1000 μL micropipettes ( 1 each)
2) iCheck® Quantitative Tester ( with zero calibration film )
3) Centrifuge ( 7000 rev / min, or higher)
4) Timer
4. Preparation before testing and calibration of the instrument
1) Restore the quick check card, buffer and sample to be tested to room temperature for 30 minutes before use.
2) Equipment zero point calibration: Insert the zero point calibration piece into the quantitative quick tester measurement room, and click “Zero point correctionâ€.
3) Equipment calibration: Insert the M1 electronic calibration card into the quantitative quick tester card slot, and click “Read Calibrationâ€. The electronic calibration card must not be removed during the measurement.
4) According to the number of samples, open the package, take out the quick check card and M1 micro-hole, and place the M1 micro-hole on the micro-hole holder.
5. Sample processing and testing
Milk, milk powder
1) A 1 mL aliquot of milk (or after reconstitution of milk) samples in 1.5 mL centrifuge tube (if not detected immediately, the milk stored refrigerated, prevent corruption caused by failure or pollution), 7000 rpm / min, centrifuged for 10 minutes.
2 ) Add 75 μL of the dilution buffer CSB-4 to the M1 microwell, then add 75 μL of the lower layer skim milk sample, pipette 5 times to mix, and take 100 μL to the sample hole of the quick test card.
3 ) After reacting for 15 minutes at 37 °C (±2 °C ) , immediately put the quick check card into the quantitative quick tester and click the “ analyze †reading, which is the actual concentration of the sample.
6. Result determination
1 ) Detection range: 0-2ppb, the limit of quantification is 0.25ppb.
2 ) This test card is a quantitative quick check card. If there is no red line on the control line ( C ), it indicates that the quick check card is invalid.
3 ) The aflatoxin M 1 measured value is greater than 0.4 ppb , please re-measure it once. If it is still greater than 0.4ppb , please use the instrument method for confirmation analysis.
7. Precautions
1) The quick check card is used once during the warranty period. Store the refrigerator at 2-8 °C until it is balanced to room temperature before use.
2) Beware of the moisture in the quick check card, the aluminum foil packaging bag is damaged or the quick check card is damp, the test card should not be used.
3) Try not to touch the white film in the center of the quick check card.
4) Avoid direct sunlight during all incubations.
5) The small bag in the aluminum foil packaging bag is a desiccant and should not be taken into account.
6) Exceeding the shelf life cannot be used.
