When these three methods are used, the ionic strength and pH of the soaked section buffer, the temperature and time of heating all affect the antigen retrieval effect.

1, the application range and choice of pH

The pH of the antigen retrieval solution is very important, and the effective antigen retrieval pH is more important than the chemical composition of the repair solution. The same repair solution gradually increases in intensity with increasing pH, but the optimal pH range is 6.0-10.0. The best antigen-repairing solution currently recognized is citrate buffer pH 6.0 and EDTA buffer pH 8.0. As a general-purpose repair solution, the alkaline pH of the repair solution is more effective than the acid, and for the old archived tissue that has been fixed for a long time, the acidic pH repair solution is superior to the alkaline repair solution.

2, the best temperature should be selected when antigen repair

The temperature of 70-90 ° C can be denatured to unfixed proteins, but formalin-fixed proteins must be at a temperature above 92 ° C to denature them. The results show that a temperature of 92-98 ° C is suitable, 95 ° C is the best.

3, antigen repair fluid must follow the law of natural cooling, otherwise the effect is not good or the purpose of antigen repair is not achieved

After the duration of antigen retrieval, remove it and let it cool down slowly at room temperature. Never use ice or cold water to cool it for time. This is because when the antigenic protein molecular chains in the high temperature are separated from other binding or bonding, there is a natural environment that allows them to naturally relax, and as the temperature decreases, they slowly return to their original form and configuration.

4, try to use a sufficient amount of antigen repair solution

The liquid used for antigen retrieval must be sufficient to prevent the slices from drying out. When a large amount of antigen retrieval solution is applied, due to its large amount, the boiling time of the liquid can be delayed, and the amount of the microwave radiation of the slice is increased, and the antigen retrieval will be thorough.
